What do you want to edit? Do you want to change a variable? Add a variable? Assuming it's the latter, you could just use the -append flag.
For example
x = rand(100,1);
save('MyData.mat','x'); % initial save
y = rand(100,1);
save('MyData.mat','y','-append'); % add a variable
