How to identify Defects on a Image?.

Hi, I am developing a Defect Identification software using Visual Studio. But I am not succeed. I heard using matlab it's possible. What I want to do is there will be a reference image which taken by the camera. I want to compare the images reference and the captured image. My Images are not registered because we have manual feeding system. Now I want to compare the images and identifying the defect. Is this possible to do with matlab?. If any one knows then kindly give me some suggestion how to do. I am expecting the answers as soon as possible.
