How to decide the pixel distance, gray levels and orientation(angle) before calculating GLCM from gray scale images?

Hey guys, i am currently working on finding a method to classify diffracted images belonging to different type of cells using GLCM and then calculating few features from GLCM. As there are many factors to be considered before we can calculate GLCM. How do we decide on the pixel distance , angle and gray level scaling before calculating GLCM? For example,in the below attached pdf,there are 2 different types of cells on page 1 and page 2. With our naked eye we can easily differentiate but in order to classify many cells like this i want to calculate the glcm and extract few features for each type of cells and then may be use a classifier to identify them. But how do i know which combination(pixel distance, gray levels and angle) will fetch good result?
