I did an operation on an image and found out the output figure. Now, i want to use that figure and do operations on it. How should i read it using imread or any other way possible.

Give an example. You almost never want to do an operation on a figure (the gray background with axes, an image, tick marks, annotations, etc.). You almost always want to do operations directly on the image itself.
If you really want to save a figure window as an image and read it back in with imread, then use export_fig(), followed by imread(). http://matlab.wikia.com/wiki/FAQ#How_do_I_save_my_figure.2C_axes.2C_or_image.3F_I.27m_having_trouble_with_the_built_in_MATLAB_functions.

Then don't do anything on the figure. Do it on the image array. See my tutorials in the File Exchange: http://www.mathworks.com/matlabcentral/fileexchange/?term=authorid%3A31862
